Separation of Terfenadine Enantiomers by High Performance Liquid Chromatography Using β-Cyclodextrin as Chiral Mobile Phase Additive

摘要 以 β 环糊精为手性流动相添加剂 ,于C1 8反相柱上建立了特非那丁手性对映体的高效液相色谱拆分方法。探讨了β 环糊精浓度、有机修饰剂种类及含量、流动相pH等因素对特非那丁对映体拆分的影响 ,结果表明 :当流动相为高氯酸钠溶液 (0 .0 8mol L)∶乙醇∶二乙胺 (75∶2 5∶0 .5 ) ,pH 6.2 ,含β 环糊精浓度为 2 5mmol The enantioselective resolution of terfenadine enantiomers by reversed phase high performance liquid chromatography with β-cyclodextrin as a chiral mobile phase additive is described. A C 18 reversed phase column was used as separation stationary phase. The effect of some factors such as concentration of ethanol and β-cyclodextrin and the pH of mobile phase on the resolution was investigated. The results showed that nearly baseline separation of terfenadine enantiomers was obtained as mobile phase were composed of NaClO 4 (0.08 mol/L)∶ethanol∶diethylamine (75∶25∶0.5) with 25 mmol/L β-cyclodextrin, at pH 6.2.
