摘要
目的 探讨基因重组人类肿瘤坏死因子 α(rh TNF- α)和 bcl- 2反义寡核苷酸 (ASPO)对 HL- 6 0细胞增殖与凋亡的作用 ,进一步认识肿瘤坏死因子 α(TNF- α)和 bcl- 2两者调控肿瘤细胞凋亡的作用及相互联系。 方法 单独和联合应用 rh TNF- α和 bcl- 2 ASPO处理 HL- 6 0细胞 ,通过 MTT法检测 HL- 6 0细胞生长和存活特性变化 ;原位细胞凋亡检测和流式细胞技术检测细胞凋亡 ;应用 RT- PCR、免疫组织化学检测癌基因 bcl- 2表达的改变。 结果 rh TNF- α和 bcl- 2 ASPO的联合应用明显增强对 HL- 6 0细胞的增殖抑制 ;细胞凋亡率明显升高 ;癌基因bcl- 2在 m RNA和蛋白表达明显下降。 结论 TNF和 bcl- 2 ASPO在诱导 HL- 6 0细胞凋亡中可能存在协同作用 。
Objective\ To investigate the effects of reformed human tumor necrosis factor\|α(rhTNF\|α) and bcl\|2 antisense sephosphorthioate oligodeoxynucleotides(ASPO) on acute myeloid leukemic cell line HL\|60, to learn more about the relationship between TNF\|α and bcl\|2 on apoptosis of the hematological malignancies.\ Methods\ The effects of rhTNF\|α and bcl\|2 ASPO on the proliferation and apoptosis of leukemic cell line HL\|60 were studied by using MTT assays, flow cytometer(FCM) technique and in situ cell apoptosis detection.\ To detect the variation of bcl\|2 oncogene expression through RT\|PCR and immunohistochemical staining.\ Results\ Both rhTNF\|α and bcl\|2 ASPO inhibit the proliferation and induce the apoptosis of HL\|60, the combinative therapy enhance the effects signigicantly.\ The sharply down\|regulation of bcl\|2 oncogene expression can also be observed.\ Conclusion\ Cytokine TNF\|α in combination with bcl\|2 ASPO probably have synergistic effects on apoptosis of hematological malignancies, so the regimens including cytokine and antisense oligodeoxynucleotides may provide a new way for treatment of hematological malignancies.\;
