摘要
目的 探讨慢性HBV感染者外周血树突状细胞 (DC)和T细胞对激活剂体外应答的功能。方法 用三色荧光染色法 ,通过流式细胞仪检测静息期和经激活剂激活后CD11c+DC和CD3 +/CD4+/CD69+T细胞的百分率。结果 与接种过乙型肝炎疫苗、血中抗HBs阳性健康者相比 ,感染者组CD11c+型DC及其表达CD80和CD40共刺激分子的阳性细胞百分率 ,以及经CD2 /CD2R强激活剂刺激后CD69+T细胞百分率等三项指标均呈明显低下的状态。结论 慢性HBV感染者血中DC和T细胞对激活剂的体外刺激 ,均表现不同程度的应答功能低下或缺陷 ,本研究涉及的三项细胞免疫检测方法有利于深入了解机体对病原体应答的能力 ,其结果有助于阐明发病机理 。
Objective To study the immune function of peripheral blood dendritic cells(DCs) and T cells in patients with chronic hepatitis B virus(HBV) infection. Methods Whole peripheral blood, collected from patients with chronic HBV infection and healthy donors, was stimulated with HBsAg or CD2/CD2R. Then three-color staining flow cytometry was used to detect the membrane marks of DCs and T cells. Results The percentages of CD11c + DCs, CD80 + DCs, CD40 + DCs, and CD3 +/CD4 +/CD69 +T cells were all significantly lower in patients than in control. These results indicate that the immune functions of DCs and T cells in patients with chronic HBV infection were decreased in response to stimulator. Conclusion Membrane marks, such as CD11c, CD80, CD40 of DCs and CD69 of T cells, can assist to comprehend immune response function. These results are very useful in clarification the pathology and specially in selection of the immune therapy methods in chronic hepatitis B.
