摘要 以皮皮虾壳(甲壳素)为原料,再用NaOH溶液进行处理使甲壳素脱去乙酰基制备壳聚糖。采用单因素试验的方法寻找NaOH溶液的质量分数、处理时间、处理温度以及甲壳素和NaOH溶液的投料比对脱乙酰基的影响,以探索出壳聚糖最优的制备条件。结果表明,壳聚糖最优的制备条件:将甲壳素按投料比1:300(甲壳素质量和NaOH液的体积比)加入到45%的NaOH溶液中,在水浴温度为85℃的水浴锅中加热3小时,用自来水清洗至中性,室温下晾干。制备的壳聚糖含水量为9.0%,灰分含量为0.4%,脱乙酰度为84.1%。 Taking Pippi shrimp shell(chitin) as raw material, the chitosan was prepared by dislodging the acetyl group with chitin by handling with sodium hydroxide solution. The effects of the mass fraction of sodium hydroxide solution,the processing time, the processing temperature and the ratio of chitin and sodium hydroxide solution on the deacetylation were investigated by single factor experiment to explore the optimal preparation conditions of chitosan. The results showed that chitosan was prepared by soaking chitin in mass fraction 45% NaOH at a feeding ratio of 1:300(chitin mass and NaOH volume ratio), heated at 85℃ for 3 h, then washed the chitosan to neutral and dry at room temperature.The prepared chitosan was with 9.0% of moisture content, 0.4% of ash content, the degree of deacety-lation at 84.1%.
