摘要 华蒲公英具有较强的环境适应能力,是植物抗逆性研究的优良材料。通过多因素分析种子最佳消毒时间,萌发、增殖和生根培养基的最适激素浓度。结果显示,最佳消毒方案为75%乙醇消毒30 s+无菌水冲洗2次+0. 1%升汞消毒10 min+无菌水冲洗5次,最佳萌发培养基的激素选择是6-BA 0. 2 mg/L、GA3 1. 0 mg/L,最佳增殖培养基的激素选择为NAA 0. 1 mg/L、6-BA 0. 5 mg/L,最佳生根培养基的激素选择是NAA 1. 0 mg/L。通过实验研究,筛选华蒲公英组培不同阶段的培养基配方,建立华蒲公英组培体系。 Taraxacum borealisinense Kitam.isa perennial herb with the same source of medicine and food,belonging to Taraxacum,Compositae.In this study,we used Taraxacum borealisinense seeds as experimental materials and established the tissue culture system by optimizing culture conditions in each stage.The results showed that the optimumdisinfection method was that the material was disinfected with75%ethanol for 30 s and rinsed with sterile water for twice,and then disinfected with 0.1%mercuric chloridefor 10 min and rinsed with sterile water for four times.The optimal germination medium was MS medium supplemented with0.2 mg/L6-BA and 1.0 mg/L GA3.The optimal proliferation medium was MS medium supplemented with 0.1 mg/L NAA and 0.5 mg/L6-BA.The optimal rooting medium was MS medium supplemented with 1.0 mg/L NAA.Through experiments,the medium formulations of different stages of tissue culture were screened to establish a system of dandelion tissue culture.
