摘要
目的比较黄芩、白芍对牙周炎小鼠血清IgG和IgG1水平的影响,探讨黄芩和白芍对牙周炎的免疫干预和治疗作用机制。方法 64只清洁级12周龄雄性昆明小鼠随机分成4组,每组16只。牙周炎组、黄芩治疗组、白芍治疗组采用丝线结扎上颌第一磨牙并在结扎部位涂菌4周,建立小鼠实验性牙周炎模型,于建模后第5周分别以蒸馏水、黄芩水提物和白芍水提物灌胃。各组动物于建模后第4、6、8、10周各处死4只。制做牙体牙周组织联合切片,显微镜下观察牙周组织的病理变化。酶联免疫吸附测定(ELISA)法检测血清中抗体IgG和IgG1的水平。结果牙周炎组可见牙周组织破坏明显。较牙周炎组相比,黄芩治疗组和白芍治疗组牙周组织炎症程度均明显减轻。与牙周炎组相比,黄芩治疗组和白芍治疗组血清IgG水平在6、8和10周逐步升高(P<0.01)。黄芩治疗组在各时间点的血清IgG水平较白芍治疗组提高显著(P<0.01)。黄芩治疗组和白芍治疗组在6、8和10周IgG1抗体水平均显著升高(P<0.05)。黄芩治疗组血清IgG1水平在各时间点均较白芍治疗组升高显著(P<0.05)。结论黄芩、白芍可以增强抗体IgG的水平和强化Th2细胞反应,达到治疗小鼠牙周炎的效果;黄芩对小鼠牙周炎的治疗效果优于白芍。
Objective To explore the effect of Scutellaria baicalensis Georgi and Paeoniae alba Radix on experimental periodontitis in mice,and the serum levels of IgG and IgG1 were compared. Methods Sixty- four SPF 12- week- old male Kunming mice were randomly divided into four groups. Experimental periodontitis group( P) Scutellaria baicalensis Georgi treatment group( SG) and Paeoniae alba Radix treatment group( RG) were prepared by wrapping silk ligature and inoculating with putative periodontopathic bacteria,the mice were gavaged with distilled water the aqueous extraction of Scutellaria baicalensis Georgi and the aqueous extraction of Paeoniae alba Radix each from week 5. Four mice in each group were sacrificed at each time point at the end of 4、6、8 and 10 weeks. The histopathological changes of periodontal tissue were observed with H&E staining. The levels of serum IgG and IgG1 were measured by ELISA. Results A serious inflammatory response was observed in the experimental periodontitis group. However,in SG and RG,the inflammation in periodontal tissue significantly decreased. The levels of serum IgG in SG and RG at 6,8,10 weeks significantly increased( P < 0. 01); The level of serum IgG in SG was significantly higher than that of RG( P < 0. 01). The levels of serum IgG1 in SG and RG at 6,8,10 weeks were significantly increased( P < 0. 05); The level of serum IgG1 in SG was significantly higher than that of RG( P < 0. 05). Conclusions Both Scutellaria baicalensis Georgi and Paeoniae alba Radix showed therapeutic effects on periodontitis in mouse models by the mechanism of strengthening serum IgG and Th2 response. The Scutellaria baicalensis Georgi was more effective on experimental periodontitis than Paeoniae alba Radix.
