黑色素瘤抗原基因在肝癌细胞系中的表达 被引量:1

Expression of melanoma antigen gene in cell lines originated from hepatocellular carcinoma cells

摘要 目的 研究黑色素瘤抗原 (MAGE)基因在肝癌 (HCC)细胞系中的表达 ,为应用MAGE抗原肽作为疫苗治疗肝癌的体外实验筛选靶细胞。方法 分别用逆转录 聚合酶链反应(RT PCR)和免疫组织化学方法对MAGE 1和MAGE 3mRNA及MAGE 1蛋白在 8种HCC细胞系中的表达进行检测 ,用微量淋巴细胞毒法测定HCC细胞系的主要组织相容性复合物 (MHC)Ⅰ类分子的表达。结果  6种HCC细胞系MAGE 1基因mRNA阳性表达 ,2种MAGE 3阳性 ;有 5种细胞系MAGE 1基因蛋白阳性。BEL740 5HLA A2 4阳性 ,HLE和HpG2HLA A2阳性。 结论 MAGE基因在HCC细胞系中有较高的表达率 ,HLE和BEL740 5可作为阳性靶细胞 ,其他细胞系可作为相应的对照细胞 ,应用于用MAGE抗原肽疫苗治疗HCC的体外实验研究中。 Objective To study the expression of me la noma antigen (MAGE) gene in hepatocelllular carcinoma (HCC) cell lines and to sc reen target cells for the study of using MAGE peptide as a new vaccine for immun otherapy for HCC.Methods The mRNA of MAGE-1 and AGE-3 a nd the protein of MAGE-1 in 8 HCC cell lines was detected using RT-PCR and imm unohistochemical staining.The type of major histocompatibility complex (MHC) Ⅰ was studied with microcytotoxicity.Results MAGE-1 mRNA w as expressed in 6 cell lines,and MAGE-3 mRNA in 2 cell lines;MAGE-1 protein e xpression was positive in 5 cell lines.The expression of HLA-A24 was detected in the cell line of BEL7405 and the expression of HLA-A2 was found in the cell lines of HLE and HpG2.Conclusion MAGE genes were expresse d at high percentage in HCC cell lines.HLE and BEL7405 as candidate positive ce ll lines and other cell lines as various kinds of control cells could be used in the study of using MAGE peptide as a new vaccine in immunotherapy for HCC.
