摘要
建立经皮冠状动脉成形术 ( PTCA)后再狭窄动物模型是研究再狭窄机制的基础 ,根据国人冠状动脉的解剖特点 ,用直径 2 .5 mm球囊导管 ,损伤兔髂动脉内膜 ,2 8d后 ,组织经 HE染色、VanGieson胶原染色、α-肌动蛋白免疫组化实验 ,通过图像分析系统分析 ,测定狭窄指数和增殖指数。结果表明 ,此动物模型能较好地模拟 PTCA后血管再狭窄的病理过程 ,证实平滑肌细胞的增生是引起再狭窄的主要原因。
Establishment of Rabbit iliac artery restenosis model after percutaneous transluminal coronary angioplasty (PTCA)is the base for research the mechanism of restenosis. According to the character of coronary artery of Chinese people, we established an animal model by using 2.5-mm balloon catheter to denude endothelium in rabbits iliac artery. Twenty eight days after operation, the tissue sections were stained with HE (hemalaune hematoxylin and eosin) and van Gieson for morphologic examinations. Smooth muscle cells(SMC) were indicated immunohistologically with an antibody against α-acting .The indexes of stenosis and proliferation were determined by image analysis system. The result showed that the model can simulate the pathologic procedure of restenosis after PTCA and confirmed that the proliferation of SMC is the mainly cause of restenosis.
