兔眼碱、酸、热烧伤后蛋白抗原性变化的研究 被引量:14

摘要 目的 了解角膜碱、酸、热烧伤后有无新的角膜变性蛋白产生并检测其抗原性。方法 利用SDS PAGE电泳技术和Western印迹实验分离角膜蛋白并确定其是否具有抗原性。结果 碱烧伤后 4周的角膜蛋白同正常角膜蛋白相比 ,可见在 2 3 5kda、2 5kda、3 5kda左右出现 3条新的蛋白带 ,其中 2 3 5kda和 3 5kda的蛋白能够刺激机体产生特异性的抗体并与之结合。热烧伤后 4周可见在 14 5kda左右出现l条新的蛋白带 ,不能刺激机体产生特异性的抗体。酸烧伤后 4周未出现新的蛋白带。结论 在角膜碱烧伤后的损伤机制中有变性抗原与抗体的病理性免疫反应参与 ,并持续较长时间存在。 Objective To explore the change of corneal protein and determine if there is any new denatured protein and detect their antigenicity following alkali burn,acid burn and thermal burn.Methods The extracted corneal protein was separated by SDS PAGE and analyzed by Western blot to determine its antigenicity.Results There were three bands of new protein with the molecular mass 23.5 kda,25 kda and 35 kda separated in the cornea of alkali burned for 4 weeks.Two specific antibodies against the corneal protein of 23.5 kda and 35 kda were detected.There was one band of new protein with the molecular mass 14.5 kda separated in the cornea of thermal burned for 4 weeks,but no specific antibodies against it.There was no new protein separated in the cornea of acid burned for 4 weeks.Conclusion In the mechanism of the alkali burn this kind of pathological immunoreaction about denatured antigen and its specific antibody plays important operation,and exists for long time.This kind of pathological immunoreaction doesn't exist in the mechanism of the thermal burn and acid burn.
