摘要
目的:建立正常人胆囊组织基因表达目录。方法:采用含17000cDNA克隆的基因微矩阵与2例正常胆囊之cDNA杂交,取基因表达平均值,分析胆囊的基因表达谱。结果:正常胆囊有11047条基因表达,其前10位高表达基因与平滑肌收缩及物质转运有关。在前200位高表达的基因中,离子通道和转运蛋白、蛋白翻译和合成类以及发育相关类基因占多数。在胆囊所有表达的基因中,12、2、21号染色体表达的数目最多。另外,21条胆石病候选基因在胆囊中有表达,其中主要为脂类代谢相关基因。两条胆囊特异性表达基因:NPC1L1、SLC17A2。结论:胆囊高表达的基因可能与胆囊收缩和胆汁浓缩有关,胆囊不仅参与机体的消化功能,还参与调节脂类代谢平衡。
Objective:To elucidate the gene expression profile in normal gallbladder tissue.Method:A cDNA array representing17000cDNA clusters was used to hybridize with cDNA of2normal gallbladder tissues specimens,and the average expression abundance was adopted to analyse the gene expression profile of normal gallbladder tissue.Results:There were a total of11047genes expressed in the normal gallbladder,and the first10highly expressed genes were related to the function of smooth muscle contraction and materials transport.In the first200highly expressed genes,those related to ion channel and prorein transport,protein translation and synthesis,and development were listed in the top3positions.The top3chromosomes were12,2and21respetively when all the gene expression profile was elucidated.In the meanwhile,21gallstone candidate susceptibility genes were expressed in the normal gallbladder,most of them were lipid metabolism related genes.Inaddition,another2genes:NPC1L1and SLC17A2,were found peculiarly expressed in the normal gallbladder.Conclusion:The genes highly expressed in the normal gallbladder might be related to gallbladder contraction and bile concentration;and the gallbladder takes part in both the digestion and lipid homeostasis.
