摘要
目的 :观察自发性高血压大鼠 (SHR)高血压产生不同时期一氧化氮合酶 (NOS)各亚型在肾脏中的分布及变化。方法 :应用免疫组织化学方法用NOS亚型的 3种特异抗体 (NOS1、NOS2和NOS3)分别对SHR高血压产生各时期肾脏NOS各亚型 (bNOS、eNOS、iNOS)的表达进行检测。结果 :在正常对照组WistarKyoto(WKY)中 ,NOS1主要在球旁斑细胞中有强阳性反应 ,而在其它部位如内髓收集管、内髓升支粗段、肾小球和血管中未发现阳性反应。在第 4周、第 9周和第 14周染色无明显变化 ;与WKY比较 ,SHR在第 4周时bNOS表达明显增多 ,第 9周bNOS阳性细胞数依然增多 ,但不如第 4周时明显 ,在第 14周时WKY与SHR未见明显区别。而两种大鼠肾脏中eNOS和iNOS的变化无显著差异。结论 :SHR不同时期一氧化氮合酶 (NOS)各亚型在肾脏中的分布及变化可能参与高血压发病机制 ,球旁斑细胞内生成的NO参与调节球旁区的血管舒张反应。
Objective:To investigate the expression of each isoforms of nitricoxide synthase in different period of kidney in spontaneously hypertensive rats (SHR).Method:Three special antibodies NOS1,NOS2,NOS3 were used to determine the distribution of each isoforms of nitricoxide synthase respectively in different period of kidney by immunohistochemistry method.Results:There was positive response of NOS 1 in macula cells of periglomerular while the other parts such as the collecting duct of kidney medulla, pachysegment part of ascending branch, renal glomeruli and vassals were negtive response in normal contrast of Wister Kyoto(WKY), there were no significance during 4W,9W 14W; the expression of bNOS was significant higher positive response in 4W in SHR than that in WKY ,and there were no significance during 9W and 14W between SHR and WKY.The expression of eNOS and iNOS were no significance between SHR and WKY.Conclusion:The distribution of each isoforms of nitricoxide synthase in different period of kidney in SHR has relation with pathogenesis of hypertension ,and NO generated from macula cells of periglomerular regulate the vasodilatatio response.
