系统性红斑狼疮患者外周血单个核细胞c-kit受体表达的研究

Expression of cell kit on peripheral blood monocytes of patients with systemic lupus erythematosus

目的 探讨系统性红斑狼疮 (SLE)患者外周血单个核细胞 (PBMC)中c kit受体的表达及其临床意义。方法 采用免疫荧光标记、流式细胞仪检侧SLE患者PBMC的c kit受体蛋白(CD117)表达 ,反转录 聚合酶链式反应 (RT PCR)检测其c kitmRNA的表达水平 ,并分别与正常人比较。结果 SLE患者PBMC中c kit受体及其mRNA表达水平与正常组相比显著增高 (P <0 0 5 ) ;活动期与非活动期相比显著增高 (P <0 0 5 ) ;非活动期SLE患者PBMC的c kit受体蛋白表达与正常人差异无显著性 (P >0 0 5 )。PBMC的c kit受体蛋白表达与其SLEDAI显著相关 (P <0 0 1)。结论 SLE患者PBMCc kit受体蛋白水平及其mRNA表达水平均显著高于正常人 ,且与疾病活动呈正相关 ,c kit受体可能在SLE的病程变化中起着重要作用。

Objective To determine the level of cell KIT (c kit) of peripheral blood monocytes (PBMCs) in patients with recent onset systemic lupus erythematosus (SLE),and analyze the relationship between the c kit of PBMCs and clinical parameters.Methods The level of c kit of PBMCs in patients with SLE was estimated with flow cytometry,and c kit mRNA was examined by reverse transcript polymerase chain reaction (RT PCR).Results The c kit level of PBMCs in SLE patients ( n =47) was significantly higher than that in healthy controls ( n =20),the c kit level of PBMCs in active patients ( n =27) was significantly higher than that in inactive patients ( n =20).There was no significant difference between active and inactive patients ( P >0 05).Conclusion C kit level may be more closely related to the clinical parameters in SLE patients,which may reflect the clinical status of SLE patients.
