摘要
目的:观察TRAIL对结肠癌细胞株SW480的细胞毒作用.方法:采用MTT法检测TRAIL对结肠癌SW480细胞存活分数的影响;TUNEL法检测TRAIL对结肠癌SW480细胞凋亡率的影响.结果:TRAIL能有效地杀伤结肠癌SW480细胞,MTT法显示1000μg/L的TRAIL作用24hSW480细胞存活分数仅为18.7%.TUNEL法均显示0,50,150,500,1500,5000μg/LTRAIL蛋白诱导结肠癌SW480细胞的凋亡率分别为6.7%,29.4%,42.8%,50.8%,84.6%和87.4%;500μg/LTRAIL蛋白于0,6,12,18,24和30h诱导结肠癌SW480细胞的凋亡率分别为7.8%,21.4%,41.8%,60.6%,73.8%和77.3%.TRAIL对结肠癌SW480细胞的作用存在较好的量效关系和时效关系.结论:TRAIL能通过诱导细胞凋亡的方式杀伤SW480细胞,有可能成为有效的抗结肠癌新药.
AIM:To study cytotoxic effect of TRAIL on colon cancer cell line SW480. METHODS:The viability of SW480 cells was measured by MTT assay and the apoptotic rate was determined by TUNEL method. RESULTS:Results of TUNEL and MTT assay showed that TRAIL had high antitumor activity in a time- and concentration- dependent manner. Survival rate of SW480 cells after TRAIL (1 000 μg/L) treatment was 18.7 %. The apoptotic rates of SW480 cells after exposure TRAIL at concentration of 0,50, 150,500,1 500 and 5 000 μg/L were 6.7 %,29.4 %,42.8 %, 50.8 %,84.6 % and 87.4 %, respectively. The apoptotic rates of SW480 cells after treatment with 500 μg/L TRAIL for 0,6,12,18,24 and 30 hours were 7.8 %,21.4 %,41.8 %, 60.6 %,73.8 % and 77.3 %, respectively. CONCLUSION:The results demonstrate that TRAIL can induce apoptosis and destruction of SW480 cells, and is a potential new cytotoxic drug in cancer therapy.
