Construction of pUCM-T vector of human mature form transforming growth factor beta 1

目的 构建人成熟型TGF β1基因T克隆载体 ,为进一步研究人成熟型TGF β1的原核表达和功能打下基础。方法 应用RT PCR技术从人外周血单个核细胞扩增人成熟型TGF β1编码区cDNA基因 (336bp) ,并将其克隆至pUCM T载体 ,经限制性核酸内切酶EcoRI,HindIII的酶谱分析和DNA测序分析证实。结果 成功构建人TGF β1/T载体 ,可进一步用于基因表达和表达产物的功能研究。结论 人TGF β1/T载体的构建 ,为进一步研究其基因表达和功能打下基础 。

Objective In order to provide an essential preparation for obtaining a large quantity of recombinant human mature form transforming growth factor beta 1 (TGF β 1), the cDNA of human mature form TGF β 1 was cloned and the pUCM T vector of mature form TGF β 1 was constructed. Methods According to the nuclear acid sequence of human mature form TGF β 1, the gene was cloned with RT PCR from human peripheral blood mononuclear cell. Then the gene was subcloned into the pUCM T vector, and the sequence of humam mature form TGF β 1 was proved to be correct by sequencing. Results The humam mature form TGF β 1 cDNA was successfully inserted into pUCM T vector, and the sequence of human mature form TGF β 1 was correct. Conclusion The construction of human mature form TGF β 1 provides the basis for the further study.
