摘要
目的 :探讨Fas/FasL和C myc基因表达与隐睾生殖细胞凋亡的关系 ,以及C myc与Fas/FasL介导的细胞凋亡途径之间的关系。方法 :采用 2 2d龄SD雄性大鼠建立单侧隐睾模型 ,运用生物素 dUTP/酶标亲和素测定法检测睾丸生殖细胞凋亡 ,运用免疫组织化学SP法检测Fas/Fasl和C myc基因表达。结果 :①与对侧睾丸相比 ,术后第 3天隐睾侧睾丸发生凋亡的生殖细胞无显著增加 (P >0 .0 5 ) ,Fas/FasL和C myc基因表达亦无显著增加 (P >0 .0 5 ) ;第 7天隐睾侧睾丸发生凋亡的生殖细胞显著增加 (P <0 .0 1) ;Fas/FasL和C myc基因表达均有显著增加 (P <0 .0 1)。②实验性隐睾术后第 3天和第 7天 ,C myc基因表达与相应Fas/FasL基因表达间均存在着正相关关系。结论 :手术诱导的隐睾生殖细胞凋亡增加 ;Fas/FasL和C myc基因表达上调可能是生殖细胞发生凋亡的重要原因之一 ;隐睾生殖细胞凋亡可能是通过与C
Purpose:To investigate Fas&FasL、C myc expression and testicular germ cell apoptosis in experimental cryptorchidism.Method:Immature rats (22 days old Sprague Dawley) were subjected to unilateral cryptorchidism. Using terminal deoxynucleotidyl transferase mediated d UTP nick end Labeling (TUNEL) for the detection of germ cell apoptosis; Using immunohistochemical SP method for the detection of Fas&FasL、C myc expression. Result:①At the third day after operation, as compared to the contraposition control, neither the number of apoptotic germ cell, nor the augment of Fas&FasL、C myc expression was highly notable (P> 0.05 ) in the cryptorchidism. The number of apoptotic germ cell increased markedly (P< 0.01 ) and the augment of Fas&FasL、C myc expression was highly notable (P< 0.01 ) in the cryptorchidism as compared to the contraposition control at the seventh day after operation. ②There is a considerable correlation coefficient between the expression of C myc and the respective expression of Fas&FasL at the third day and the sevConclusion:Enhanced expression of Fas&FasL、C myc might be involved in germ cell apoptosis in cryptorchidism; testicular germ cell apoptosis in experimental cryptorchidism occurs by Fas FasL mediated apoptosis in association with C myc.
