摘要
目的探讨不同温度对人舌鳞癌细胞系(Tca-8113)表皮生长因子受体复合物内化的影响。方法用氯胺T法标记125I-hEGF。采用放射受体结合分析法比较在37℃和4℃时,125I-hEGF细胞表面结合的量和内化的量。结果37℃时,在10min后受体复合物内化超过细胞表面结合的量。4℃时,内化量一直维持较低水平。结论4℃时,配体结合反应稳定,适宜配体结合分析实验。EGFR复合物内化现象的研究,为临床筛选EGFR单克隆抗体进行治疗提供了实验依据。
Objective To investigate the effect of different temperatures on internalization of epidermal growth factor receptor complexes in human tongue squamous carcinoma cells(Tca-8113).Methods Human epidermal growth factor r(hEGF)was labeled with 125 I according to Chloramine-T method.The quantity of surface binding and internalization of 125 I-hEGF in Tca-8113cells at4℃and at37℃was compared respectively with radiobinding assay.Results At37℃,the amount of EGFR complexes internalization exceeded that of surface binding after10min.At4℃,the level of interlization maintained very low.Conclusion The study indicated that the surface binding of 125 I-hEGF at4℃was steadier than that at37℃.The study on phenomenon of EGFR complexes internalization provide the experimental basis for selecting anti-EGFR monoclonal antibody in clinic.
