羟基磷灰石/磷酸三钙复合材料成骨细胞载体的体外实验 被引量:22

摘要 目的:研究人骨髓基质细胞(humanbonemarrowstromalcells,hBMSC)在复合材料的表面生长和增殖情况,为成骨细胞载体研究和材料表面改性提供直接依据。方法:进行原代人骨髓基质细胞培养,传代至第3代后,将hBMSC和羟基磷灰石(hydroxyapatite,HA)/磷酸三钙(tricalciumphosphate,TCP)进行混合培养,采用倒置显微镜、荧光显微镜和扫描电镜观察形态,MTT测定细胞增殖情况,测定碱性磷酸酶了解分化情况。结果:hBMSC在HA/TCP复合材料上生长、增殖良好,并可见细胞向材料孔内生长,对碱性磷酸酶无明显影响。结论:HA/TCP复合材料是良好的骨组织工程的细胞载体,为进一步临床应用提供了基础。 AIM:To study the effect of HA/TCP matrix scaffold as osteogenic cells carrier in vitro.METHODS:Human bone marrow stromal cells primary culture were performed, at the third passage, cells were cultured on HA/TCP matrix scaffold, fluorescence microscope and scaning microscope were used to observe the cells,proliferation test was performed by MTT method at 3rd and 6th day, the supernant liquid were checked for alkaline phosphatase.RESULTS:Human bone marrow stromal cells showed good biocompatibility with HA/TCP matrix scaffold, the scaffold promoted human bone marrow stromal cells proliferation slightly.CONCLUSION:HA/TCP matrix hydroxyapatite, tricalcium phosphate, human bone marrow stromal cells scaffold is a good osteogenic cells carrier.
