摘要
运用办公用扫描仪和一个自编软件进行定量分析。该方法能将一次扫描成像的一列(或一排)多个斑点同时转换为以斑点的灰度积分值为纵坐标像素列位置为横坐标的峰形图,并给出灰度积分值和斑点的Rf值。研究了该方法对硅胶薄层板、滤纸、打印纸上所点碱性品红、萘酚绿B、果绿、普鲁氏蓝、Cu2+-K4Fe(CN)6棕红5种色斑进行定量分析的可行性。结果表明各色斑的灰度积分值与点样体积、浓度、样品绝对量成正比。不同点样介质上色斑的灵敏度和精密度不同。
A software for utilizing officeuse scanners in qualitative and quantitative analysis of thinlayer chromatography was developed. The software can transfer a series of color spots in a line scanned simultaneously into a peakform graph, in which Ycoordinate represents the grayscale of spots and Xcoordinate represents the positions of spots. TheRf of a peak and the integral grayscale value of a peak area were put out. The relationships between the spots' grayscale integral value of five colors: basic fuchsin, fruit green, naphthol green B, KFe and Cu2+-K4Fe(CN)6 spoted on silicagel thinlayers, filter papers and printer papers with different concentration, volume and area were investigated. The spots' grayscale integral value is proportional to their volume or concentration of the color solution or the total amount of a color compound of the solution. The same spots spoted on different medium have different sensitivity and precision.
