P53,C-met及C-erbB-2癌基因蛋白在胃癌中的表达及分布特征 被引量:2

摘要 目的 通过对胃癌标本中 P5 3,C- met及 C- erb B- 2癌基因蛋白的表达分析 ,探讨其在胃癌发生中的作用。 方法 用免疫组化方法检测了 6 0例胃癌标本中 P5 3,C- met及 C- erb B- 2的表达及分布。 结果  P5 3,C- met及 C- erb B- 2在胃癌中的表达分别为 5 1.7%、70 .0 %、5 0 .0 %;胃癌旁正常粘膜上皮的阳性率分别为 1.6 %、16 .7%、8.3%,与肿瘤区相比较有显著差异 (P<0 .0 1) ,P5 3阳性细胞的染色部位在细胞核内 ,C- met及 C- erb B- 2阳性细胞的染色部位在胞浆及 /或胞膜 ,C- met癌旁正常粘膜区与肿瘤区染色强度比较有差异 (P<0 .0 5 ) ,C- erb B- 2在肿瘤移行区与肿瘤区比较有差异 (P<0 .0 5 ) ,P5 3及 C- erb B- 2阳性表达与肿瘤的分化程度有关。 结论  P5 3,C- met及 C-erb B- 2与胃癌的发生、发展密切相关 ,P5 3及 C- erb B- 2在肿瘤分化过程中可能起重要作用。 Objective To investigate the role of P53, C-met and C-erbB-2 in gastric carcinogenesis. Methods Expression and distributive feature of P53, c-met and C-erbB-2 in 60 cases of human gastric cancer were detected with immunohistochemical staining. Result The positive rate of expression was respectively 51.7% for P53, 70.0% for C-met and 50.0% for c-erbB-2; and that in the gastric mucomembrane nearby carcinomas was respectively 1.6% for P53, 16.7% for C-met, 8.3% for c-erbB-2. The different expressions of P53, c-met and c-erbB-2 oncoproteins in carcinoma and non-carcinoma tissues were very obvious(P<0 01). The positive cells of P53 oncoprotein appeared in nuclei of tumor cells, while the positive cells of c-met and c-erbB-2 oncoproteins appeared in the membrane or plasma of tumor cells. There was difference (P<0 05) in the c-met staining indensity of mucomembrane nearby carcinomas vs tumor region, there was difference(P<0 05) in the c-erbB-2 staining indensity of translated region vs tumor region. The positive expressions of P53 and c-erbB-2 were related to the differentiation of tumour tissues. Conclusion P53 ,c-met and c-erbB-2 were closely correlated with occurrence and devolopment of gastric carcinoma, and the expression of P53 and c-erbB may contribute to tumor differentiation..
