ox-LDL作用下丘脑、垂体条件培养液对内皮细胞MDA代谢和ICAM-1表达的影响 被引量:1

摘要 目的 :观察ox LDL作用下丘脑垂体后条件培养液对内皮细胞MDA代谢和ICAM 1表达的变化 ,探讨下丘脑血管中枢对AS形成的影响。方法 :按下丘脑 (H组 )、下丘脑 +垂体 (HP组 )、ox LDL作用下丘脑 (ox H组 )、ox LDL作用下丘脑 +垂体 (ox HP组 )和对照组 5组分别收集条件培养液 ,培养正常及受损内皮细胞。收集上清液检测MDA含量 ,细胞用免疫细胞化学法检测ICAM 1表达。结果 :MDA和ICAM 1的变化 :4个实验组的条件培养基对正常内皮细胞MDA的代谢和ICAM 1的表达都有下调作用 (P <0 .0 5 ) ;而对受损内皮细胞MDA的代谢和ICAM 1的表达有所不同 ,ox H组和ox HP组与H组和HP组相比 ,两者的下调作用明显减弱 (P <0 .0 1) ;与同组的正常内皮细胞相比 ,有明显加重脂质过氧化作用 (P <0 .0 1)。结论 :ox LDL作用下丘脑垂体后条件培养液对正常内皮细胞起抗损伤作用 。 Objective:To observe the changes of MDA metabolism and ICAM 1 expression of endothelial cells cultured with the condition media of hypothalamus pituitary and to investigate the influence of hypothalamic vessels's center on pathogenesis of AS.Methods:The condition media of hypothalamus(H), hypothalamus+pituitary(HP), hypothalamus treated with ox LDL(ox LDL), hypothalamus+pituitary treated with ox LDL(oxHP),and control were collected respectively.The normal and damaged EC were cultured separately with the condition media. The supernatants were collected respectively for detecting the content of MDA and the cells were fixed for immunocytochemical staining to show the expression of ICAM 1. Results: The changes of MDA and ICAM 1:The condition media of all experimental groups down regulated the metabolism of LP0 and expression of ICAM 1 of normal EC( P <0.05), especially in the HP and ox HP groups; while the down regulation effect on ox LDL were significant depression as compared with those of H and HP group ( P <0.01);There was a significant increase in lipoperoxidation as compared with normal endothelial cells of the same group ( P <0.01).Conclusion:The condition media of hypothalamus pituitary treated with ox LDL shows anti damage effects on the normal EC and does little of anti damage effects on damaged EC.
