摘要 综述了谷子的组织培养与基因工程研究进展。谷子组培对培养基要求不严,可选用MS、N_6、L_3为基本养基,琼脂的浓度为0.7%~0.8%,蔗糖的浓度是3%,pH值5.8。2,4-D是主要的诱导愈伤组织激素,愈伤的继代可选用如下激素IAA,NAA,BA,LBA等。外植体可用种子,幼牙,枝条,未成熟的种子或胚,幼穗。基因枪转化中常用载体为pBI_(121) and pAHC_2常用启动子为35S启动子与ubi启动子,选择标记以Kn为主,GUS为报告基因。谷子基因转化的参数正逐渐优化。 Tissue cultrue and trans-gene of millet were reviewed. Millet tissue didn' t need strict medium. Callus can be initiated on MS, N6, L3 medium. The medium was solidified with 0.7% - 0. 8% agar,3% sucrose,and pH 5.8.2,4-D was the most efficient hormone. Subculture of the callus was promoted by IAA,NAA,BA,LBA and so on.Seeds,roots,shoots,immature embryos and in some case immature seeds can be used as explant. pB1121 and pAHC25 plasmid were used in millet gene gun bom-bardmemt. Camv35S and ubiquitin promoter were constructed and comparied. Kanamycin-resistance gene and GUS gene was used as markers gene.
