摘要 目的 探讨甲基叔丁基醚 (MTBE)对小鼠肝细胞生长周期动力学及细胞DNA含量的影响。方法 昆明种小鼠经呼吸道静式染毒 ,MTBE浓度为 10 8 0 ,14 4 0 0及 4968 0mg/m3,每日 1次 ,每次 4h ,连续染毒 2 0d。流式细胞仪技术研究MTBE对小鼠肝细胞周期、细胞增殖及细胞DNA含量的结果。结果 MTBE各染毒组小鼠肝细胞的G0 /G1,S ,G2 /M期细胞数及增殖指数PI与对照组相比差异未见显著性 (P >0 0 5 ) ;MTBE 14 4 0 0与 4968 0mg/m3染毒组小鼠肝细胞DNA指数低于对照组 (P <0 0 5 )。结论 MTBE对小鼠肝细胞周期、细胞增殖无显著影响 。 Objective To study the effects of methyl tertiary butyl ether(MTBE)on cell cycle kinetics and DNA content of hepatocytes in mice.Methods Mice of Kunming strain were exposed to MTBE at concentrations of 108 0 mg/m 3,1 440 0 mg/m 3 and 4 968 0 mg/m 3 for four hours daily in consecutive 20 days via respiratory tract.Flow cytometry technique was used to study cell cycle,cell proliferation and DNA content in hepatocytes.Results No significant increase in cell number and proliferation index(PI)were found in the phases of G 0/G 1,S and G 2/M of hepatocytes in all exposed mice,as compared with the controls(P>0 05).DNA index lowered significantly in hepatocytes of the mice exposed to 1 440 0 mg/m 3 and 4 968 0 mg/m 3 of MTBE,as compared with the controls(P<0 05).Conclusions MTBE had no significant effect on cell cycle and cell proliferation in hepatocytes,but could cause decrease in their DNA content.
