摘要
对花生与其近缘野生种间细胞融合及培养方法进行研究。将花生品种鲁花14号的体细胞胚和近缘野生种A.cannadacii的胚性愈伤组织分离得到的原生质体,用PEG方法融合后,培养在添加1mg/LNAA和5mg/LBAP的改良MS液体培养基中。培养4d后,细胞开始分裂。之后部分细胞继续分裂,并形成小愈伤组织。
The protoplast fusion and culture in peanut and its related species were studied. Protoplasts isolated from somatic embryos of peanut cv. Luhua No.14 and embryogenic calli of Arachis cannadacii were fused by PEG method, and then cultured in the modified liquid MS medium containing 1 mg/L NAA and 5 mg/L BAP. The first cell division occurred within 4 days after fusion. And some of the cells further divided and developed into visible small calli.
