摘要
目的 探讨银屑病细胞凋亡的机制。方法 用原位末端标记(TUNEL)技术检测银屑病皮损及经银屑病患者白细胞刺激的培养角质形成细胞的凋亡情况。透射电镜观察其超微结构的变化。结果 银屑病组凋亡表达为89.31±27.23,对照组为18.72±8.74(P<0.01)。经银屑病患者白细胞刺激的角质形成细胞凋亡表达为0.9585±0.3421,较各对照组明显增高(P值均<0.01)。超微结构显示其细胞增生活跃,细胞间粘连疏松。角质形成细胞内见凋亡改变。结论 推测凋亡增多伴随细胞过度增殖是机体维持细胞动力学稳定的一种保护性机制。凋亡增多可能与银屑病患者外周血白细胞参与的诱导有关。
Objective To investigate the mechanism of apoptosis in psoriasis. Methods The apoptosis in psoriasic lesions and keratinocytes stimulated by psoriatic leucocytes was detected by TUNEL asssy. The ultrastructural changes were observed by transmission electron microscopy. Results The apoptosis expression was 89.31 ± 27.23 in the psoriatic lesions and 18.72 ± 8.74 in the control group (P<0.01). The apoptosis expression in the keratinocytes stimulated by psoriatic leucocytes was 0.9585± 0.3421 .significantly higher than in the control group (P<0.01). Ultrastructurally, there were neuclear hyperplasia of keratinocytes and loose connection between keratinocytes. Conclusion The increase of apoptosis and hyperplasia of keratinocytes may be one of protecting mechanisms to maintain stability of cell dynamics of body. Overexpression of apoptosis may be related with induction by peripheral blood leucocytes in the patients with psoriasis.
