果糖二磷酸钠镁对大鼠心肌缺血再灌注损伤的影响 被引量:2

摘要 目的 探讨果糖二磷酸钠镁 (FDPM)对大鼠心肌缺血再灌注损伤的干预作用及其机制。方法  5 0只Wistar大鼠随机分为 5组 :假手术组 (Sham) ,模型对照组 (Control) ,果糖二磷酸钠镁组 (FDPM) ,1,6 二磷酸果糖组 (FDP)和硫酸镁组 (Magsul)。采用左冠状动脉下穿线 ,拉紧丝线引起心肌缺血 ,放松丝线给予再灌注的方法建立心肌缺血再灌注损伤动物模型。放松丝线前5min开始给予相应药物 ,持续到再灌注 10min。记录不同时相点血液动力学变化以及再灌注室性心律失常情况。实验终点时分别取血和心肌标本测定丙二醛、心肌坏死面积等指标。结果 与对照组相比 ,3个用药组均能明显减少心肌坏死面积 ,改善心脏功能 ,抗氧自由基损伤 ,降低梗塞区钙离子含量 (P <0 .0 5 ) ;Magsul和FDPM有防止再灌注心律失常作用 ,FDP则没有 ;在改善心肌再灌注后收缩功能方面 ,FDP和FDPM疗效优于Magsul。结论 FDPM能够干预大鼠心肌缺血再灌注损伤 ,很可能是通过FDP和Magsul协同作用而产生 ;FDPM疗效虽不明显优于FDP或Magsul,但具有防止再灌注心律失常和改善心肌收缩功能的双重作用。 Objective To explore the beneficial effects of sodium magnesium fructose diphosphate(FDPM) on myocardial ischemia/reperfusion injury (I/R I) in rats.Methods Fifty Wistar rats were randomly divided into five groups:control,sham, FDPM,FDP(fructose 1,6 diphosphate) and Magsul (magnesium sulfate).Models were made by drawing then releasing the suture advanced around the left coronary artery in rats inducing 35 minutes ischemia and 90 minutes reperfusion.Agents were administered during the period of 5 minutes before and 10 minutes after releasing.LVSP,LVEDP and ±dp/dtmax were measured and recorded by pressure transducer.Ventricular arrhythmias during reperfusion were continually recorded and scored.Blood and heart samples were withdrawn at 90 minutes reperfusion for assay of CK and LDH activities;Heart samples were used to determine the area of infarction,activities of ATPase and SOD,contents of calcium ion and MDA.Results Compared with control,the three agents all reduced the zone of myocardial necrosis,decreased LVEDP,improved the activities of ATPase and SOD,lowered the activities CK and LDH,as well as reduced contents of MDA and calcium ion,however,there was no a statistically significant difference between them.FDPM and FDP were superior to Magsul in improving heart function,on the other hand , in preventing ventricular arrhythmias after reperfusion, Magsul and FDPM were superior to FDP ( P <0.05) .Conclusion FDPM can attenuate myocardial I/R I in rats,which is achieved possibly by partial synergetic action of FDP and Magsul,but these three agents' effects are similar.
