经戊二醛与环氧氯丙烷处理后猪瓣的形态与组织结构变化 被引量:2

摘要 目的 :研究戊二醛 (glutaraldehyde ,GA)与环氧氯丙烷 (epoxychloropropan ,EC)处理猪动脉瓣对其形态及组织结构的影响 .方法 :将宰杀 1h的猪主动脉瓣 4 0枚 ,平均分成 4组 ,分别置入对照组 :4℃的 30mL·L-1GA保存液中 ;GA组 :4℃的 5mL·L-1GA液保存 ;EC改性组 :经 4℃的 5mL·L-1GA液保存 4 8h后 ,移入 4℃的 2 0mL·L-1EC液保存 4 8h ,再用 4℃的 5mL·L-1GA液保存 ;EC处理组 :用 4℃的 2 0mL·L-1EC液保存 .各组在处理后 2mo观察大体形态 ,显微和超微结构 ,测定组织稳定性 ,组织含水量 ,保存 1a后测定组织断裂强度 .结果 :对照组 ,GA组 ,EC改性组的纤维形态结构无差别 ,EC处理组胶原纤维出现局灶性松乱和溶解现象 ;对照组 ,GA组和EC改性组 3组间热皱缩温度无明显差别 (P>0 .0 5 ) ,但分别明显高于EC处理组 (P <0 .0 0 1) ;4组间组织含水量无明显差别 (P >0 .0 5 ) ;4组间组织断裂强度无明显差异 (P >0 .0 5 ) .但EC改性的GA瓣组织结构的稳定性和抗钙化优于其他组 .结论 :EC改性的GA瓣较GA瓣能保持瓣膜结构的完整性并提高瓣膜组织的稳定性 ,具有更好的抗张强度和抗钙化能力 . AIM: To study the effects of glutaraldehyde (GA) and epoxy chloropropane (EC) on porcine aortic valves morphologic features and tissue structure. METHODS: Fourty porcine aortic valves were divided into four groups and were treated with different density of GA or EC and the valves treated with 3% GA were used as control group. In GA group, the density of GA was 0.5%, in EC group, the density of EC was 2% and in EC modified group, the density of GA were 0.5% and 2% and the density of EC was 2%. After two months to one year preservation, the stability, water content and breached strength were observed. The morphologic features were also observed by light and electron microscope. RESULTS: There were no difference in fiber structure among the control group, EC modified group and GA group. In EC group, the focal derangement and dissolution of fibers were observed. There were no difference of heat shrinking temperature among the control group, EC modified group and GA group ( P >0.05), but it was significantly lower in EC group ( P <0.001). There were no difference of water content and breached strength among the four groups ( P >0.05). CONCLUTION: EC chemical modification of GA treated porcine aortic valves can effectively preserve the structure and improve the tissue stability and integrity of valves and enhance the tension of porcine aortic valves and ability against calcification.
