摘要
目的 建立晶状体连续发育模型,检测Pax-6基因在小鼠晶状体发育的不同阶段的表达情况,探讨该基因在小鼠晶状体发育中的作用。 方法 利用RT-PCR法和Western blot法,检测Pax-6在小鼠晶状体发育不同阶段中的表达状况。 结果 在鼠胚9.5 d(E 9.5)即可检测到Pax-6的表达,在其连续发育阶段的E12.5,E17.5及新生10、20、60 d均可检测到Pax-6的表达。Western blot法同样发现在体外培养的原代细胞,传第1、第2及第3代的晶状体上皮细胞(LECs)中均能检测到Pax-6蛋白水平的表达。 结论 小鼠晶状体发育的各个阶段均具有Pax-6基因,该基因的正常表达是晶状体发育的必要条件。
Objective To study gene expression of Pax-6 in the different stages of lens development in mouse model. Methods The expression of Pax-6 gene in the different stages of lens development was demonstrated by RT-PCR and Western blots. Results The mRNA of Pax-6 gene expression was positive in mouse embryo at 9. 5 day ( E9. 5 ) , E12. 5 , E17. 5 ; the gene expression was also detected in the 10th day and the 20th day of new born mouse, the adult mouse at 60th day as well. The protein of Pax-6 had also positive expression in primary culture and the first, second and third passages of lens epithelial cells (LECs). Conclusions Gene of Pax-6 is positive during the different time of lens development. This result indicates that normal expression of Pax-6 gene is vital for lens development.
