双侧海马注射神经肽Y Y2受体反义基因对大鼠学习记忆影响的初步探讨 被引量:11

Effect of Intrahippocampal Infusion of Antisense Oligodeoxynucleotides to NPY Y2 Receptor on Learning and Memory in Rats

摘要 目的:探讨神经肽Y Y2(NPY Y2)受体在大鼠学习记忆中的作用。方法:双侧海马插管,注射Y2受体的反义、错义寡核苷酸或生理盐水,采用跳台法记录训练期的错误次数(学习成绩)、记忆测试期的潜伏期和错误次数(记忆成绩)。结果:注射Y2受体反义寡核苷酸组大鼠训练期的错误次数较对照组无显著性差别,而记忆测试期的潜伏期较对照组显著缩短,错误次数显著增加,提示该组大鼠记忆能力受到显著损害。结论:NPY Y2受体参与了大鼠的记忆形成过程。 Objective: To study the role of neuropeptide Y (NPY) Y2 receptor in learning and memory of rats. Methods: Anti-sense oligodeoxynucleotides (ODN) against NPY Y2 receptor mRNA were infused to block the respond of hippocampal Y2 receptor in adult rats. Control groups were infused by missense ODN or saline. And the effect of learning and memory in rats was examined by the step down test. The errors in training (learning result) and retention time and errors in retention trial period (memory result) were recorded. Results: There was no significant difference of acquisition among 3 groups. In contrast, on the retention test, there was significant difference among the Y2 receptor antisense ODN group, the saline and missense ODN control groups. Conclusion: These results provide the evidence that NPY Y2 receptor in the hippocampus takes part in the consolidation course of memory.
