肺组织粗提物促进人原发性肝癌细胞移动侵袭的研究

Effect of lung tissue extracts on migration and invasion in human hepatocellular carcinoma cells with high and low metastatic potentials

目的 探讨宿主微环境与原发性肝癌器官特异性转移的关系。方法 利用Boyden小室侵袭实验 ,分别比较从 5只C5 7BL/6小鼠提取肺 ,肝 ,脾 ,肾组织粗提物诱导高低转移潜能的人原发性肝癌细胞株MHCC97 H、MHCC97 L移动和侵袭能力。结果 肺、肝、脾、肾组织粗提物诱导高转移细胞株MHCC97 H细胞移动侵袭的细胞数分别是 ( 18.0± 0 .9)个、( 1.7± 0 .3)个、( 1.1± 0 .3)个、( 8.6± 0 .6 )个。与肝、脾、肾组织粗提物相比 ,肺组织粗提物具有显著诱导高转移潜能的人原发性肝癌细胞株MHCC97 H移动侵袭能力 ,差异有非常显著性 (P <0 .0 0 1)。肺组织粗提物诱导人肝癌细胞株MHCC97 H、MHCC97 L移动和侵袭的细胞数分别为 ( 18.0± 0 .9)个、( 6 .0± 0 .9)个 ,差异有非常显著性 (P <0 .0 0 1)。结论 肺组织粗提物中存在某种可溶性因子促进人原发性肝癌细胞的移动侵袭能力 ,宿主微环境影响原发性肝癌器官特异性转移。

Objective To investigate the effects of extracts of lung,liver,spleen,kidney on the migration and invasion of human hepatocellular carcinoma cells with different metastatic potentials.Methods The in vitro invasion assay was used to compare the ability of migration and invasion of human hepatocellular carcinoma cells,MHCC97-H and MHCC97-L,with different metastatic potentials induced by extracts of lung,liver,spleen,kidney.Results The numbers of cells in response to extracts of lung,liver,spleen,kidney in MHCC97-H were (18.0±0.9),(1.7±0.3),(1.1±0.3),(8.6±0.6),respectively.In comparison to the extracts of liver,spleen and kidney,the lung extract showed significant differences to promote the migratory and invasive ability for MHCC97-H (P<0.001).The numbers of cells in response to the lung extract were (18.0±0.9) in MHCC97-H and (6.0±0.9) in MHCC97-L (P< 0.001),respectively.Conclusion The tissue-specific factor from the extract of lung may promote the migration and invasion of the human hepatocellular carcinoma cells.Organ-specific metastasis of primary liver cancer may be mediated by host environment.
