摘要 目的:通过原代培养乳鼠皮层神经元,为脑血管疾病的研究提供实验模型。方法:酶消化法原代培养出生1天Wistar乳鼠的皮层神经元,利用免疫细胞化学和免疫荧光方法进行鉴定。结果:培养的细胞贴壁生长,胞体饱满,突起较长。免疫细胞化学染色显示神经元特异性烯醇化酶阳性细胞达85%,免疫荧光显示胶质细胞酸性蛋白阳性细胞达15%。结论:酶消化法原代培养乳鼠皮层神经元是一种可靠、稳定的获得神经元的方法。 Objective To establish an experimental model of primary culture of cortical neurons of newborn rat and find a good way to the further investigation of cerebrovascular diseases.Method Primary culture of cortical neurons of newbo rn rat was performed by the method of enzyme digestion.Evaluation of neurons was made by both immunocytochemical and immunofluores-cence techniques.Re sults Cultured neurons were stick to the wall of the cultivation plate when they were growing.The neurons were satiety and with long apophysis.Immunocytochemical staining showed85%NSE(neuro-specific enolase)positive staining cells.Meanwhile,immunofluorescence test indicated about 15%GFAP(glial fibriliary acidic protein)positive staining cells.Conclu sion Primary culture of cortical neurons of new born rat by the method of enzyme digestion is a reliable way to obtain neurons and with high stability.
