顺铂抑制豚鼠耳蜗外毛细胞钙激活的钾电流

摘要 目的 研究顺铂对豚鼠耳蜗外毛细胞钙激活的钾电流作用。方法 豚鼠被迅速断头处死 ,暴露耳蜗 ,在解剖显微镜下去除耳蜗侧壁 ,经胰蛋白酶消化并轻轻吹打 ,以获得单离的豚鼠耳蜗外毛细胞。静置后在倒置相差显微镜下观察。应用膜片钳制技术从单离的豚鼠耳蜗外毛细胞获得钙激活的钾通道记录 ,并观察顺铂对它的作用。结果 结果表明该通道活动受到细胞外浓度为 10 -5mol·L-1的顺铂的抑制。 OBJECTIVE The effects of cisplatin on K Ca currents were investigated in dissociated cochlear outer hair cells of the guinea pig. METHODS Guinea pig was decapitated and the temporal bone was quickly removed. Then the lateral wall of the cochlea was removed. The organ of Corti were dissected out under a dissection microscope. Single cells were obtained by digestion with trypsin and gentle mechanical trituration. Patch clamp technique were used to record K Ca current from isolated guinea pig cochlear outer hair cells and determine the effect of cisplatin on it.RESULTS The results showed that the activation of K Ca channels were inhibited by extracellular cisplatin in concentration of 10 -5 mol·L -1 .CONCLUSION This finding demonstrates that extracellular cisplatin depress K Ca current in the guinea pig cochlear outer hair cells.
