摘要
目的 :研究槐果碱对感染 CVB3小鼠干扰素和肿瘤坏死因子活性的影响。方法 :制备 CVB3病毒性心肌炎小鼠模型 ,用 MTT比色分析法检测小鼠脾细胞 IFN-α与 TNF的诱生量以及血清中 型干扰素 (IFN- )和TNF水平。结果 :槐果碱 5 0 mg· (kg· d) - 1 皮下注射可提高正常小鼠血清 TNF含量。与正常对照组比较 ,病毒对照组 TNF活性明显升高 ,IFN无明显变化。槐果碱 5 0 m g· (kg· d) - 1 和 2 5 mg· (kg· d) - 1 两个剂量灌胃均明显升高 CVB3感染小鼠血清和脾细胞诱生的 IFN-α活性。槐果碱两种途径、四个剂量给药均明显降低感染小鼠血清中TNF活性 ,其中槐果碱 2 5 mg· (kg· d) - 1灌胃和槐果碱 30 m g· (kg· d) - 1皮下注射还明显降低脾细胞中的 TNF诱生量。结论 :槐果碱升高 CVB3感染小鼠的 IFN活性 ,降低 TNF的活性 ,可能是其体内抗
Objective:To study the effects of Sophocarpine on activities of IFN and TNF in murine model with Coxsackie virus B 3 induced viral myocarditis.Methods:The activities of IFN α,TNF produced by splenic cell and serum IFN Ⅰ,TNF in murine model were measured by MTT assay.All data of experimental groups and blank control group were collected and compared statistically.Results:The activity of serum TNF in healthy group treated with Sophocarpine 50mg·(kg·d) -1 sc was significantly increased in contrast to that in healthy control group received normal saline.The activity of TNF in murine model with Coxsackie virus B 3 induced myocarditis was significantly increased compared with that in healthy control group.No significant differences were found in the activity of IFN between the mouse model group with Coxsackie virus B 3 induced myocarditis and the healthy control groups.The activity of IFN α in serum and produced by spelenic cells in the mouse model group treated with Sophocarpine 50mg·(kg·d) -1 ig and 25mg·(kg·d) -1 ig was significantly increased compared with that in the blank control group.The activity of serum TNF in mouse model group treated with Sophocarpine was significantly decreased compared with that in the blank control group.The activity of TNF produced by splenic cell in the mouse model group treated with Sophocarpine 25mg·(kg·d) -1 ig and 30mg·(kg·d) -1 sc was significantly decreased compared with that in the blank control group.Conclusions:The activity of IFN increases significantly,but the TNF activity decreases significantly in the mouse model group treated with Sophocarpine, as compared with that of the blank control group.
