针药结合治疗学习记忆障碍的机制——对东莨菪碱所致获得性学习记忆障碍大鼠海马NO和NOS的影响及其相关性 被引量:7

摘要 目的 探讨 NO与 N OS与大鼠学习记忆障碍的相关性 ,以揭示临床针药结合治疗影响学习记忆功能的可能机制。方法 以东莨菪碱造成学习记忆获得性障碍模型 ,分别给以尼莫地平、针刺、针药结合治疗后 ,进行跳台、迷宫测试 ,并将所测大鼠迷宫登陆时间与大鼠海马组织 NO、NOS的含量及活性进行相关性分析 ,比较针药结合治疗与单纯针刺治疗对东莨菪碱所致大鼠学习记忆障碍的改善作用。结果 针刺组 N O含量 (1.30 0± 0 .14 μm ol/g)和 NOS活性 (0 .0 64± 0 .0 0 7u/m g)显著高于尼莫地平组 (0 .80 8± 0 .2 3μmol/g,0 .0 5 6± 0 .0 12 u/m g)和针药组 (0 .70 5± 0 .5 2 μm ol/g,0 .0 47± 0 .0 0 5 u/mg) (P<0 .0 5 ) ,针刺组学习记忆能力改善显著低于尼莫地平组、针药组。结论 针药结合治疗优于单纯针刺组 ,能更有效的抑制 NOS活性 。 Objective To study the correlation between NO and NOS and learning\|memory disturbance in rats, and further to explore the possible mechanism of combining acupuncture and medicine in the treatment of it. Methods 10 rats in every group were modeled to be with learning memory disturbance by scopolamine, and then treated with nimodipine, single acupuncture, and combining acupuncture and medicine respectively. Jumping platform and labyrinth tests were conducted. The landing period and the contents of NO and NOS in the rats' hippocampi were compared correlatively. The improvements of learning memory capacity in every group were compared. Results The content of NO(1.300±0.14 μmol/g) and activity of NOS(0.064±0.007 u/mg) were significantly lower in single acupuncture group than in nimodipine group(0.808± 0.23 μmol/g,0.056±0.012 u/mg) and combining acupuncture and medicine group(0.705±0.52 μmol/g,0.047± 0.005 u/mg) ( P <0.05). The learning memory disturbance was improved obviously less in single acupuncture group than in nimodipine group and combining acupuncture and medicine group. Conclusion Combing acupuncture and medicine has better effects than single acupuncture in inhibiting NOS activity and thus reduce the damage of neurons.
