电针对局灶性脑缺血大鼠脑组织NO及NOS的影响 被引量:13

Influence of Electroacupuncture at Governor Vessel on NO and NOS in Brain Tissue in Rats with Focal Cerebral Ischemia

摘要 目的 研究电针对局灶性脑缺血后脑组织 N O含量及 3种亚型 N OS表达的影响。方法 采用改良大脑中动脉线栓法造成大鼠局灶性脑缺血模型 ,电针百会、大椎 ,应用硝酸还原酶法测定 NO含量及免疫组织化学技术检测脑组织 3种亚型NOS(n NOS、i NOS、e NOS)的表达 ,观察局灶性脑缺血后 3种亚型 NOS表达。结果 督脉电针能减少局灶性脑缺血后脑组织中 NO含量 (13.12± 3.15μm ol/L ) ,与缺血组比较 P <0 .0 1;督脉电针组缺血侧脑组织 i N OS、n N OS表达 (13.0 1± 1.32 ,32 .12± 1.5 6)显著低于缺血组 (P<0 .0 1) ,e NOS表达 (2 9.2 1± 3.0 1)显著高于缺血组 (P<0 .0 1)。结论 督脉电针能上调e N OS表达 ,同时下调局灶性脑缺血所致 n NOS、i NOS的异常表达 ,从而减少总 NO的生成量 ,降低缺血引起的毒性作用 。 Objective To study the influence of electroacupuncture at governor vessel on the content of NO and the activities of the three subtypes of NOS in the brain tissues. Methods Modified embolism method in the cerebral middle artery was employed to make focal ischemia in rats; point selection: Dazhui(GV 14) and Baihui(GV 20) nitric acid reductase method was used to detect the content of NO, and immunohistochemical method was used to detect the expressions of three subtypes of NOS (nNOS, iNOS and eNOS). Results Compared with the ischemia group, electroacupuncutre could remarkably reduce the content of NO ( P <0.01), reduce the expressions of nNOS and iNOS in the affected side of brain ( P <0.01), and increase the eNOS expression ( P <0.01}. Conclusion Electroacupuncture at governor vessel can increase eNOS expression, and reduce the expressions of nNOS and iNOS to decrease the production of NO, as a result, to reduce the ischemic injury and protect the brain tissues.
