PD-L1在口腔鳞状细胞癌及癌旁中表达情况的研究

Expression of PD-L1 in oral squamous cell carcinoma and adjacent tissues

目的:研究程序性死亡受体配体1(PD-L1)在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)中的表达情况。方法:采用免疫组化(IHC)、荧光定量逆转录聚合酶链式反应(RT-qPCR)和蛋白质印迹法(WB)检测PD-L1在OSCC癌及癌旁组织中的表达情况。结果:IHC结果显示PD-L1在OSCC患者癌组织中阳性率为69.3%(43/62),显著高于癌旁组织17.7%(11/62)。RT-qPCR结果显示PD-L1相关mRNA在OSCC患者癌组织中表达显著高于癌旁组织(3.2±0.3)倍。WB结果显示PD-L1蛋白在OSCC癌组织中表达显著高于癌旁组织(2.1±0.2)倍。结论:PD-L1在OSCC患者组织中高表达,有可能参与了OSCC肿瘤细胞的免疫逃逸,有希望成为临床上的治疗靶点。

Objective:To investigate the expression of programmed death receptor ligand 1(PD-L1)in oral squamous cell carcinoma(OSCC).Methods:Immunohistochemistry(IHC),fluorescence quantitative reverse transcription polymerase chain reaction(RT-qPCR)and Western Blotting(WB)were used to detect PD-L1 in cancer and adjacent tissues.Results:IHC showed that the positive rate of PD-L1 in cancer tissues of OSCC patients was 69.3%(43/62),which was significantly higher than that in adjacent tissues 11.7%(11/62).RT-qPCR showed that the expression of PD-L1 related mRNA in cancer tissues was significantly higher than that in adjacent tissues(3.2±0.3 folds).WB showed that the protein expression of PD-L1 in cancer tissues was significantly higher than that in adjacent tissues(2.1±0.2 folds).Conclusion:The expression of PD-L1 is high in OSCC patients,which may be involved in the immune escape of OSCC tumor cells,and it is hopeful to become a clinical therapeutic target.
