摘要
目的:探讨LASP1(LIM and SH3 protein1)基因在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC中的表达水平及其预后相关性。方法:分别下载TCGA数据库中的OSCC队列数据(n=374)及GEO数据库中的GSE42743数据集(n=103)。比较LASP1基因在OSCC组和正常对照组中的表达水平,分析LASP1与OSCC患者预后的关系。此外,利用基因集富集分析方法探讨LASP1基因在OSCC中可能的作用机制。结果:LASP1基因在OSCC组中的表达水平高于对照组,差异具有统计学意义(P<0. 001)。在OSCC患者中,LASP1的表达水平与肿瘤分期呈负相关(P=0. 014),且OSCC高表达组的总生存率明显差于低表达组(Log-rank P<0. 001)。Cox回归分析结果提示LASP1基因高表达是OSCC患者预后不良的独立预测因素(P=0. 021)。基因富集分析提示,LASP1可能与黏着斑、ECM受体相互作用、Notch信号通路及癌症相关通路等生物学功能有关(P<0. 05,FDR<25%)。结论:LASP1基因在OSCC患者中的表达与OSCC患者的进展相关,可作为OSCC预后评估的一种标记物。
Objective:To explore the association between the expression of LIM and SH3 protein1(LASP1)gene and prognosis in patients with oral squamous cell carcinoma(OSCC).Methods:The OSCC cohort data from TCGA database and GSE42743 dataset from GEO database were downloaded.LASP1 expression in OSCC group and control group were compared,and the association of LASP1 with clinical features and prognosis were analyzed.Moreover,gene set enrichment analysis was performed to identify the potential mechanism of LASP1 in OSCC.Results:The expression of LASP1 was higher in OSCC than in control groups(P<0.001).In patients with OSCC,LASP1 expression was positively correlated with tumor stage(P=0.014),and the overall survival rate of high LASP1 group was significantly poorer than that of low LASP1 group(Log-rank P<0.001).Additional Cox regression analysis indicated that high expression of LASP1 was an independent predictor for poor prognosis of OSCC patients(P=0.021).Gene set enrichment analysis showed that LASP1 may be associated with focal adhesion,ECM_receptor interaction,pathways in cancer,notch signaling pathway.Conclusion:High LASP1 expression is associated with the progression of OSCC,and LASP1 might be used as a potential biomarker for prognostic evaluation of OSCC.
