基于生物信息学分析RPS19基因在肾透明细胞癌中的表达及预后意义 被引量:3

Expression and prognostic significance of RPS19 in renal clear cell carcinoma based on bioinformatics analysis

摘要 目的:通过生物大数据分析核糖体蛋白S19(RPS19)在肾透明细胞癌(ccRCC)中的表达及预后意义。方法:利用Oncomine数据库检索并筛选出RPS19的信息,得到RPS19在ccRCC的表达情况,使用GEPIA数据库分析RPS19的表达与ccRCC患者在TCGA数据中生存率之间的关系,采用LinkedOmics数据中的LinkFinder模块分析RPS19与ccRCC临床病理特征的关系。通过LinkedOmics进行RPS19在TCGA KIRC数据中的表达相关性分析,并进一步通过Metascape网站进行基因富集分析。结果:RPS19的表达水平在ccRCC组织中较正常组织明显升高。RPS19的表达与ccRCC的病理分级、T分期、N分期呈显著正相关性。高表达RPS19的患者总体生存率显著低于低表达RPS19的患者。此外,RPS19基因与RPLP2、RPS2、RPS11、PRS15、RPS16、RPS9等基因密切相关,可能通过TNF-α/NF-κB参与ccRCC的发生发展(P<0.001)。结论:RPS19在ccRCC中显著高表达,并与ccRCC的发生发展相关,大大增加其作为潜在治疗靶点的可能性。 Objective:The expression and prognostic significance of ribosomal protein S19(RPS19)in clear cell renal cell carcinoma(ccRCC)were analyzed by biological big data.Method:The information of RPS19 was retrieved and screened by Oncomine database,and the expression of RPS19 in ccRCC was obtained.The relationship between RPS19 expression and survival rate of ccRCC patients in TCGA data was analyzed by GEPIA database.The clinicopathological characteristics and correlations of RPS19 in ccRCC were analyzed by LinkedOmics,Metascape was then employed for gene enrichment analysis.Result:The expression level of RPS19 in ccRCC tissues was significantly higher than that in normal tissues.The expression of RPS19 was positively correlated with pathological grade,T stage and N stage of ccRCC.The overall survival rate of patients with high expression of RPS19 was significantly lower than that of patients with low expression of RPS19.In addition,RPS19 gene is closely related to RPLP2,RPS2,RPS11,PRS15,RPS16 and RPS9 genes,which may participate in the occurrence and development of ccRCC tumors through TNF-alpha/NF-kappa B axis(P<0.001).Conclusion:RPS19 is highly expressed in ccRCC and is associated with the occurrence and development of ccRCC,which greatly increases the possibility of RPS19 as a potential therapeutic target.
