摘要
目的:综合考察栀子药材质量。方法:采用HPLC方法建立中药栀子药材指纹图谱,色谱柱:Grace Alltima C18(4.6 mm×250 mm,5μm),流动相:乙腈-0.1%甲酸水溶液,梯度洗脱,流速为0.8 m L.min-1,检测波长为240 nm、330nm、440nm,柱温为20℃;考察了药材中栀子苷、西红花苷I、京尼平龙胆二糖苷3种指标成分TLC鉴别和水分、总灰分、酸不溶灰分、醇浸出物等药材常规理化实验项目。结果:对不同产地的栀子进行指纹分析,将所得指纹图谱分别导入《中药色谱指纹图谱相似度评价系统A版》进行谱峰匹配分析,最终成功建立了30批栀子药材的综合指纹谱库。并指认了15个共有峰;采用SPSS统计学软件,对30个不同产地的栀子常规理化实验项目测定进行聚类分析比较,结果表明,不同产地栀子药材间品质差异较大。结论:本实验为栀子药材综合质量评价体系的建立提供理论和实验依据。
Objective: To systematically evaluate the quality of Zhizi. Method: The HPLC method was used to establish the fingerprint of Zhizi. Chromatography separation was carried on Grace Alltima C18(4.6 mm ×250 mm,5 μm) column with gradient mobile phase of acetonitrile- 0.1% formic acid. UV detection wavelengths were 240 nm, 330 nm, 440 nm and the column temperature was 20 ℃ with the flow rate of 0.8 m L.min-1. Three marker compounds(geniposide, crocin I and gentian diglucoside) in samples were measured by TLC. Some physical and chemical tests(H2O, total ash, acid- insoluble ash and etoh extract) were done to investigate their qualities. Result: The fingerprints of Zhizi from different habitats were introduced into the 'similarity evaluation system for chromatographic fingerprint of TCM', and the matching of fingerprints was analyzed. Finally, HPLC fingerprints of 30 samples of Zhizi were established. 15 common peaks were selected as the fingerprint peaks in all samples. The physical and chemical parameters of Zhizi from 30 different habitats were analyzed by the cluster analysis using SPSS statistical software. There were significant differences existed in the quality of Zhizi from different production area. Conclusion: The study provides theoretical and experimental data for the systematic quality evaluation of Zhizi.
