摘要
本文报道了通过调节潮霉素和头孢霉素的使用浓度 ,在抗虫转基因水稻研究中提高筛选率和绿苗再生率。筛选时潮霉素使用浓度为 2 0~ 30mg/L ,头孢霉素为 30 0~ 40 0mg/L ;分化时潮霉素使用 15~ 2 5mg/L ,头孢霉素为 2 5 0mg/L ,可得到较高的抗性愈伤组织筛选率和绿苗分化率 ,不同水稻材料有差异。分化时绿苗假阳性率在 10 %左右可得到较大量的阳性植株和减小工作量。
In screening system of rice genetic transformation,the concentration of Hygromycine B and Cefotaxime Sodium had great effect on the frequency of screening and regeneration of transgenic calli.When the selective medium contained 20-30mg/L Hgromycine B and 300-400 mg/L Cefotaxime Sodium,the most transgenic calli were obstained in this research,while 15-25mg/L Hgromycine B and 250 mg/L Cefotaxime Sodium were used for regeneration.The proper concentration of Hgromycine B and Cefotaxime Sodium could be different with genotypes.
