摘要
目的 :探讨灯盏花素是否具有促进体外培养牛小梁细胞增殖并降低其分泌细胞外基质的作用。方法 :在体外培养的第三代牛小梁细胞中加入中药灯盏花的提取物灯盏花素 ,培养 2 4h后 ,采用放射免疫分析法测定培养液中透明质酸 (hyaluronicacid ,HA)、层粘连蛋白 (laminin ,LN)、胶原蛋白Ⅲ型 (collagen Ⅲ ,PC Ⅲ )的含量 ,同时采用噻唑蓝 (MTT)比色法测定活细胞的光密度值从而反映活细胞的数量。结果 :5mg/ml、0 .5mg/ml浓度的灯盏花素组 ,牛小梁细胞数量增多 ;5mg/ml、0 .5mg/ml浓度的灯盏花素组 ,培养液中胶原蛋白Ⅲ型的浓度降低 ,0 .0 5mg/ml浓度的灯盏花素组 ,透明质酸和层粘连蛋白的浓度降低 ;硼酸缓冲液对细胞增殖及细胞外基质浓度均无影响。结论 :灯盏花素具有促进牛小梁细胞增殖、降低牛小梁细胞分泌细胞外基质 (透明质酸、胶原蛋白Ⅲ型和层粘连蛋白 )的作用。
Objective:To investigate whether erigeron breviscapus(Vant.)(EB) can improve the concentration of bovine trabecular meshwork cells(CTCs) cultured in vitro and reduce the concentrations of hyaluronic acid(HA),laminin(LN) and protein collagen Ⅲ(PC Ⅲ).Methods:EB was added to bovine trabecular meshwork cells 3 th subcultured in vitro. After 24 hours,radio immunity was used to analyze the concentrations of HA,LN and PC Ⅲ to determine the number of active cells. Light density was used to analyze active cells with the MTT method.Results:The concentration of bovine trabecular meshwork cells increased in the 5 mg/ml and 0.5 mg/ml EB solution groups and the concentration of PC Ⅲ was simultaneously reduced. The concentrations of HA and LN decreased in the 0.05 mg/ml EB solution group. A boric acid buffer solution had no effect on the experimental results.Conclusion:Erigeron breviscapus can improve the concentration of bovine trabecular meshwork cells cultured in vitro and reduce the concentrations of hyaluronic acid,laminin and protein collagen Ⅲ.
