摘要
目的 研究内毒素血症小鼠腹腔巨噬细胞膜脂的改变及其对炎症因子产生的影响。方法 建立小鼠内毒素血症模型 ,观察内毒素 (LPS)对小鼠腹腔巨噬细胞膜脂成分、膜流动性及细胞分泌炎症因子的影响 ,并观察磷脂脂质体对细胞膜的保护作用。结果 LPS作用 1h ,巨噬细胞膜主要的磷脂成分降解(P <0 0 5 ) ;细胞膜荧光偏振度 (ρ)、微粘度 (η)和分子排列有序性系数 (γ)增加 (P <0 0 5 ) ;培养细胞上清中炎症因子血栓素B2 (TXB2 ) ,6 κ 前列腺素F1α (6 κ PGF1 α)含量明显增加 (P <0 0 5 )。经磷脂脂质体预处理后 ,膜损伤及炎症损伤减轻。结论 内毒素血症小鼠腹腔巨噬细胞膜磷脂降解 ,细胞膜从相对比较流动变为相对固化 ,与膜脂有关的炎症介质浓度增加。磷脂脂质体有修复膜磷脂结构。
Objective To investigate changes of membrane phospholipids and its effect on some inflammatory factors in mice with endotoxemia.Methods Components of phospholipids,secretion of inflammatory factors(TXB 2,6 κ PGF 1α)and the membrane fluidity were observed after mice were injected with LPS or liposome.Results 1h after the mice were injected with LPS,main phospholipids of membrane were degradated;the membrane polarizationand and microviscocity of membrane bilayer,contents of TXB 2 and 6 κ PGF 1 α in supernatant of cultured macrophages were significantly increased.Pretreated with liposome,contents of membrane phospholipids and membrane fluid were significantly increased,contents of TXB 2 and 6 κ PGF 1 α were significantly decreased. Conclusion The membrane of macrophage changes from a relatively fluid state to relatively solid one hour after being injected with LPS in mice.Phospholipid liposome could repair membrane damage,inhibit secretion of inflammation factors. [
