摘要
目的 :观察芪丹通脉片 (QDTMT)对异丙肾上腺素引起心肌损伤时其损害心肌细胞间粘附分子 1(intercellularadhe sionmolecule 1,ICAM 1)表达的影响 ,从而评价QDTMT对心肌损害中的保护作用 .方法 :SD大鼠 32只 (雄性 ) ,随机分成 4组 (模型组、正常对照组、高剂量组和低剂量组 ) ,1mo灌胃给药后 ,参考Rone等[1] 异丙肾所致心肌损害的方法改进 ,处理组腹腔注射异丙肾上腺素 4mg·kg-1·d-1,间隔 2 4h ,连续 3d ,建立异丙肾上腺素所致心肌损伤模型 ;通过免疫组化方法检测心肌ICAM 1的表达 .结果 :对照组大多数心肌的ICAM 1的表达为阴性 ,而缺血组受损心肌大多表达阳性 (85 % ) ,QDTMT作用后心肌表达ICAM 1的阳性率明显下降 (P <0 .0 1) ,以高剂量组显著 (6 3% ) .结论 :在异丙肾所致心肌损害的心肌ICAM 1表达明显增加 ,而经灌服QDTMT大鼠心肌ICAM 1表达下降 ,尤以高剂量组明显 。
AIM: To find the protective effects of Qidantongmai tablet (QDTMT) on impaired myocardium by investigating the effects of on the expression of ICAM 1 on isoproterenol impaired myocardium. METHODS: Rats were randomly divided into four groups(control, model, QDTMTmax, QDTMTmin), treated with either saline or extract of QDTMT (1 g·kg -1 ) and QDTMT (2 g·kg -1 ) respectively for one month, and then given celiac injection of isoproterenol (4 mg/kg·d -1 ) at an interval of 24 hours for three days, thus establishing the isoproterenol impaired model. The expression of ICAM 1 was detected by immunohistochemistry. RESULTS: Expression of ICAM 1 in group of control was mostly negative, but in model group, most were positive on isoproterenol impaired myocardium (85%). But after treatment with QDTMT, positive expression decreased significantly ( P <0.01), especially in the group of QDTMTmax (63%). CONCLUSION: Expression of ICAM 1 on isoproterenol impaired myocardium significantly increases and QDTMT markedly reduces the expression of ICAM 1, which indicates that QDTMT can protect isoproterenol impaired myocardium.
