b-FGF、IGF-Ⅱ、TGF-β影响兔成骨细胞体外增殖的实验研究

摘要 目的 研究碱性成纤维细胞生长因子 (b -FGF)、胰岛素样生长因子Ⅱ (IGF -Ⅱ )和转化生长因子- β(TGF- β)单独及交互联合作用 ,对兔成骨细胞体外增殖的剂量效应及时间效应。方法 采用细胞培养技术及噻唑蓝比色法 ,观察3种生长因子对兔成骨细胞体外增殖的调节作用。结果 b-FGF和IGF -Ⅱ在0.1~10.0ng/ml浓度范围内与兔成骨细胞增殖呈剂量效应关系。TGF - β 在低浓度 (0.01ng/ml)时可促进兔成骨细胞的增殖 ,随着浓度的增加 ,促增殖作用明显减弱。5d后 ,生长因子促进兔成骨细胞体外增殖作用达到最大。3种生长因子联合应用效果叠加。结论 3种生长因子均能促进兔骨髓来源成骨细胞的体外增殖 ,其作用有剂量依赖性和时间依赖性。 Objective To investigate the effects of basic fibroblast growth factor(b-FGF),insulin-like growth factor-Ⅱ(IGF-II)and transforming growth factor-β(TGF-β)on osteoblast cell proliferation of rabbit.Methods Isolated rabbit osteoblast cells were cultured in F12supplemented with b-FGF,IGF-II and TGF-β,at various concentrations and combinations.Dose-response and time-course were studied by MTT colorimetric method.Results The promotion effects of b-FGF and IGF-Ⅱincreased with the concentrations between0.1ng/ml and10.0ng/ml.TGF-βpromoted the proliferation of osteoblast cells at the dose of0.1ng/ml,while with the increasing dose,the proliferation effect decreased.The synergetic effects were achieved when the growth factors were used in combination.Conclusion The results suggest that b-FGF,IGF-II and TGF-βenhance the proliferation of osteoblast cells and the optimal combination of growth factors can increase the effect significantly.
