妥泰对沙土鼠脑缺血再灌注损伤后MDA、SOD、NO NOS及组织学的影响 被引量:1

摘要 目的 研究妥泰对脑缺血再灌注损伤的治疗作用及作用机制。方法 采用沙土鼠双侧颈总动脉结扎制成的全脑缺血模型。 2 1只动物随机分成 3组 ,即A组 :假手术 7天后组 ;B组 :缺血 5分钟再灌注 7天组 ;C组 :妥泰治疗组。测定各组沙土鼠血、脑组织的MDA、SOD、NOS含量 ,光镜电镜下观察脑组织病理及超微结构改变。结果 治疗组沙土鼠脑组织中的MDA、NO含量较缺血对照组 (即B组 )脑组织中明显降低 (P <0 0 1) ,而SOD含量明显升高 (P <0 0 1) ,治疗组 :光镜下CA1区细胞排列基本整齐 ,细胞带明显增宽 ,尼氏小体着色明显加深 ,有少量核固缩 ;电镜下 :细胞核形态较规则 ,整个膜系统基本完整 ,高密度小体及空泡明显减少 ,缺血改变明显减轻。结论 妥泰能有效抑制氧自由基的产生及毒性 。 Objective: To study the therapeutic effect of Topamax on cerebral iscvhemic reperfusion damage in gerbils and the mechanism. Methods: The whole cerebral ischemic model was made by the occlusion of bilateral carotid arteries in gerbils. Twenty-one gerbils were randomly divided into three groups: group A; sham operation group B ---ischemia for 5 min. and reperfusion for 7 days; and treatment group C with topamax. The concentrations of MDA, SOD, NO, NOS in blood plasma and brain of each group were measured. The changes of cerebral histopatholgy were observed under light and electron microsope. Results: The MDA, NO concentrations of blood plasma and brain in group C were significantly lower than those in group B(P<0.01) and the SOD concentrations in group C were obviously higher than those in group B(P<0.01). The changes of cerebral histopathology in group C were slight compared with those in group B. Conclusion: Topamax can inhibit the cytotoxic effect of oxygen free radicals on cerebral neurons in rats. So Topamax has the preventive and therapeutic effect on cerebral ischemic damage.
