摘要
目的 :小鼠转移性乳腺癌的发展与肺转移的关系。方法 :应用流式细胞仪 (Flowcytometry,FCM)动态检测小鼠移植性乳腺癌细胞DNA指数 (DNAindex ,DI)、S期细胞百分率 (S - phasefraction ,SPF)、DNA倍体 ,并分析上述3种指标与移植天数及肺转移的关系。结果 :⑴SPF随移植天数增加而减少 ,二者间呈负的直线关系 (r= -0.611,P=0.014)。⑵肺转移前后DI有显著性差异 ,肺转移前DI高于肺转移后DI。⑶小鼠移植瘤的DI与移植天数间无直线关系 ,但存在一个趋势即自移植后第10日起DI呈上升趋势直至第36天 ,而后逐渐下降至实验结束。⑷肺转移前后SPF无显著性差异。⑸DNA倍体在肺转移前多表现为二倍体 ,在肺转移后表现为异倍体 ,后期又表现为二倍体。结论 :用流式细胞仪分析小鼠移植性乳腺癌生长与肺转移的关系对临床研究有重要的意义。
Objective:To study the relationship between development of breast cancer and lung metastasis in mouse.Methods:DNA ploid,DNA index(DI)and s-phase fraction(SPF)were tested consecatively in implanted murine breast cancer cells using FCM,their relationship with the implanting days and development of lung metastasis was analyzed.Results:①SPF deˉcreased as the implanted tumors grew.That was significant in statistics(r=-0.661,P=0.014).②Higher DI was observed in groups before lung metastasis developed.(1.1913±0.2903vs0.7118±0.1185,P=0.005).③DI of implanted tumors in mouse increased from the10th day after implanting until the36th day.Then,it began to decrease gradually but no statistic significance.④Though there was no significant difference between SPF before and after lung metastasis happened,it was found that higher SPF was observed before lung metasˉtasis developed.⑤Most implanted tumors in mouse were diploid(6/8)before lung metastasis happened,while after that,they turned aneuploid and then diploid again until the experiment ended.Conclusion:It is important for clinical study to analyze the relationship between develˉopment of implanted murine breast cancer and its lung metastasis using FCM.
