摘要
对青岛文昌鱼神经胚cDNA进行测序 ,获得了 4个Sec61γ的EST ,经拼接得到编码文昌鱼Sec61γ基因全长cNDA序列并据此得到Sec61γ的氨基酸序列 .通过对Sec61γ蛋白的结构分析及其与多种脊椎动物同种蛋白的同源性进行比较 ,发现Sec61γ与人、鼠、犬及果蝇的Sec61γ ,具有较高的同源性 ,从一个侧面证明文昌鱼在进化上是位于无脊维和脊椎动物之间 ;同时说明Sec61γ基因在进化上具有较高保守性 .
By sequencing the Qingdao amphioxus neurulae cDNA library,We obtained four expressed sequence tags(ESTs) of amphioxus Sec61 γ.Compare them and then make them together into an open reading frame sequence of the cDNA of Sec61 γ.There is a high degree of similarity between amphioxus Sec61 γ and human Sec61 γ,rat Sec61γ and dog Sec61 γ.The deduced amino acid sequence was analyzed and compared with other vertebrates and invertebrates Sec61 γ proteins.The results showed that the Sec61 γ protein was conserved in evolution.Result of the transmembrane regions of all the Sec61 γ proteins also shows the conservation of Sec61 γ.It shows the amphioxus are close to vertebrates from the comparation with amino acid sequence and secondary structure of Sec61γ proteins
