摘要
目的探讨GINS2在甲状腺癌中的表达及其对甲状腺癌细胞增殖和凋亡的影响。方法通过基因干扰技术抑制甲状腺癌K1和SW579细胞中的GINS2的表达。采用Real-time PCR、Western blot检测细胞中GINS2 mRNA和蛋白的表达水平。采用MTT法和流式细胞仪检测细胞增殖、凋亡及细胞周期。结果干扰GINS2后,K1和SW579细胞GINS2mRNA和蛋白表达水平均显著低于对照组,差异具有统计学意义(P<0.01);实验组K1和SW579细胞的增殖受到显著抑制(P<0.01),凋亡率显著升高(P<0.01)。K1细胞的细胞周期发生显著改变,但SW579细胞的细胞周期未发生显著改变。结论 GINS2在甲状腺癌细胞中高表达,下调GINS2表达可抑制甲状腺癌细胞增殖并促进其凋亡。GINS2可能是甲状腺癌诊断或预后的潜在生物标志物及治疗靶点。
Objective To investigate the expression of GINS2 in thyroid cancer and its effect on the proliferation and apoptosis of thyroid cancer cells.Methods Expressions of GINS2 were inhibited in K1 and SW579 cells by using gene interference technology.Real-time PCR and Western blot were used to detect the expression of GINS2 in mRNA and protein levels in cells.MTT assay and flow cytometry were used to detect cell proliferation,apoptosis and cell cycle.Results After GINS2 was interfered,the mRNA and protein levels of GINS2 were significantly lower in K1 and SW579 cells than in control group(P<0.01).Cell proliferation was significantly inhibited(P<0.01)and apoptosis rate increased(P<0.01)in both K1 and SW579 cells.Cell cycle changed significantly in K1 cells,but not in SW579 cells.Conclusion GINS2 is highly expressed in thyroid cancer cells,and its down-regulation can inhibit the proliferation of thyroid cancer cells and promote apoptosis.GINS2 may be a potential biomarker for diagnosis or prognosis and a drug target for therapy.
