摘要
目的:探讨天门冬对衰老雌鼠卵巢沉默信息调节因2相关酶1(SIRT1)、p53水平变化。方法:以18只清洁级SD雌性大鼠为研究对象,对照组和衰老组雌鼠自由饮用自来水、天门冬组饮用天门冬水煎剂,每3 d对雌鼠进行称重,于6和72周股动脉取血处死。光镜下计数卵巢闭锁卵泡和黄体,采用qPCR检测SIRT1 mRNA,免疫组化法检测p53水平。结果:与对照组比较,衰老组和天门冬组体重逐渐增加,差异均有统计学意义(P<0.05)。与对照组比较,衰老组和天门冬组闭锁卵泡和p53蛋白明显增加,差异均有统计学意义(P<0.05)。天门冬组闭锁卵泡和p53蛋白低于衰老组,差异均有统计学意义(P<0.05)。与对照组比较,衰老组和天门冬组黄体和SIRT1 mRNA明显降低,差异均有统计学意义(P<0.05)。衰老组黄体和SIRT1 mRNA低于天门冬组,差异均有统计学意义(P<0.05)。结论:衰老可促进卵泡闭锁,黄体退化,SIRT1表达降低,p53水平增高。通过给予天门冬干预,可降低卵泡闭锁,缓解黄体退化,SIRT1表达增高,p53水平降低。
Objective:To investigate the changes of SIRT1 and p53 levels in the ovary of aging female rats.Methods:Eighteen clean SD female rats were studied.The female rats in the control and aging groups were free to drink water and the asparagus group drank the asparagus decoction.The female rats were weighed every 3 days.In 6 and 72 weeks,femoral artery blood was taken out.Ovarian atresia follicles and corpus luteum were counted under light microscope.SIRT1 mRNA was detected by qPCR and the immunohistochemical method was used to detect the level of p53.Results:Compared with the control group,the weight of the aging and asparagus groups gradually increased and the difference was statistically significant(P<0.05).Compared with the control group,the atresia follicular and p53 protein in the aging and asparagus groups increased significantly and the difference was statistically significant(P<0.05).The atresia follicular and p53 protein in the asparagus group were lower than those in the aging group and the difference was statistically significant(P<0.05).Compared with the control group,the corpus luteum and SIRT1 mRNA in the aging and asparagus groups decreased significantly and the difference was statistically significant(P<0.05).The corpus luteum and SIRT1 mRNA in the aging group were lower than that of the asparagus group.Conclusion:Aging can promote follicular atresia and luteal degeneration,lower the expression of SIRT1 and increase the level of p53.Through the intervention of asparagus,it can reduce follicular atresia,alleviate luteal degeneration,increase the expression of SIRT1 and lower the level of p53.
