摘要 目的:研究何首乌主要成分二苯乙烯苷在胃肠道的吸收。方法:通过大鼠在体胃肠灌流模型,采用酚红标记法校正肠循环液体积,HPLC法同时检测各时间点二苯乙烯苷和酚红的浓度,研究不同浓度二苯乙烯苷体内吸收情况。结果:质量浓度为60,30,10μg·mL^(-1)的二苯乙烯苷在胃内每小时的吸收百分率(PA)分别为(28.65±9.75)%,(17.04±3.98)%,(13.75±5.94)%;在小肠内的PA依次为(3.36±0.40)%,(3.36±0.79)%,(4.54±0.95)%。结论:二苯乙烯苷主要在胃内被吸收,在小肠段吸收较少。在胃酸环境下,随浓度的升高,吸收率显著增加,推测二苯乙烯苷在胃内的吸收机制为被动转运。不同浓度二苯乙烯苷在小肠内的吸收率无显著性差异,其机制可能为主动转运,还需进一步研究。 Objective:To study the gastrointestinal absorption kinetics of stilbene glucoside.Methods:The absorptions of different concentrations of stilbene glycosides were studied by rats in vivo gastrointestinal perfusion model.Phenol red labeling was used to correct the volume of intestinal circulating fluid and HPLC was used to detect the concentration of stilbene glycosides.Results:The percent absorption(PA)per hour of stilbene glucoside at concentrations of 60,30,10 μg·mL-1 were(28.65±9.75)%,(17.04±3.98)% and(13.75±5.94)% in stomach,and(3.36±0.40)%,(3.36±0.79)% and(4.54±0.95)% in intestine,respectively.Conclusion:Stilbene glycosides are mainly absorbed in stomach and less absorption in small intestine.The PA of stilbene glycosides was concentration-dependent in stomach,but there was no significant difference in intestine between different concentrations.It is hypothesized that stilbene glycoside is absorbed via passive transportion mechanism in stomach and active transportion mechanism in intestine.
