摘要
目的:观察荣筋片对SAMP6鼠血清中BMP-2水平及骨组织中BMP-2蛋白表达水平的影响,为荣筋片对抗骨质疏松症的机制研究提供新的思路。方法:选取6月龄SAMP6鼠30只,按体质量随机分为荣筋片组、空白对照组。同源对照组选取同月龄SAMR1鼠15只。荣筋片组给予3 m L药液灌胃,空白对照组给予3 m L生理盐水灌胃,均每天1次,连续12周后,摘眼球采血,ELISA法检测血清BMP-2的含量;取股骨骨髓制作涂片,免疫组化测定骨组织中BMP-2蛋白的表达。结果:荣筋片组、SAMRl对照组血清BMP-2含量与空白对照组比较,差异有统计学意义(P <0. 05);SAMRl对照组BMP-2血清含量与荣筋片组比较,差异有统计学意义(P <0. 05)。荣筋片组、SAMRl对照组骨组织BMP-2蛋白表达水平与空白对照组比较,差异有统计学意义(P <0. 05); SAMRl对照组骨组织BMP-2蛋白表达水平与荣筋片组比较,差异有统计学意义(P <0. 01)。结论:荣筋片能够提高血清BMP-2含量和骨组织蛋白表达水平,可能会改善自发性骨质疏松动物骨髓基质干细胞成骨-成脂分化BMP-2水平的异常。
Objective:By using SAMP6 and SAMR1 mice as the research object,to observe the effect of Rongjin Tablets on serum content of BMP-2 and the expression of BMP-2 protein in bone tissue.To explore the mechanism of Rongjin Tablets in preventing osteoporosis.Methods:Thirty six months old SAMP6 mice,according to the weigths,were randomized into SAMP6 Rongjin Tablets group,blank control group.Another 15 months old SAMR1 mice with resistance to rapid aging,were as the homologous comparison control group.The Rongjin Tablets group was given 3 m L solution by gavage.The blank control group was given 3 m L saline by gavage,once a day,for 12 consecutive weeks.ELISA method was used to detect BMP-2 levels in serum.Immunohistochemistry method was to test the BMP-2 protein expression in bone tissue.Results:Serum BMP-2 of blank control group was significantly lower than the other two groups(P<0.05).Serum BMP-2 of Rongjin Tablets group was significantly lower than that of the Rongjin Tablets group(P<0.05).Compared with the blank control group,the expression of BMP-2 protein was significantly lower than that of the other two groups(P<0.05).Rongjin Tablets group’s BMP-2 expression was significantly lower than that of the SAMR1 group(P<0.01).Conclusion:Rongjin Tablets can improve serum BMP-2 content and bone tissue protein expression and osteogenesis-lipophilic differentiation of BMP-2 in spontaneous osteoporosis.
